Leucine Regulates Translation Initiation of Protein Synthesis in Skeletal Muscle after Exercise

ABSTRACT
High-performance physical activity and post exercise recovery lead to significant changes in amino acid and protein metabolism in skeletal muscle. Central to these changes is an increase in the metabolism of the BCAA leucine. During exercise, muscle protein synthesis decreases together with a net increase in protein degradation and stimulation of BCAA oxidation. The decrease in protein synthesis is associated with inhibition of translation initiation factors 4E and 4G and ribosomal protein S6 under regulatory controls of intracellular insulin signaling and leucine concentrations. BCAA oxidation increases through activation of the branched-chain α-keto acid dehydrogenase (BCKDH). BCKDH activity increases with exercise, reducing plasma and intracellular leucine concentrations. After exercise, recovery of muscle protein synthesis requires dietary protein or BCAA to increase tissue levels of leucine in order to release the inhibition of the initiation factor 4 complex through activation of the protein kinase mammalian target of rapamycin (mTOR). Leucine's effect on mTOR is synergistic with insulin via the phosphoinositol 3-kinase signaling pathway. Together, insulin and leucine allow skeletal muscle to coordinate protein synthesis with physiological state and dietary intake.

    This stuff is great, soreness is way down especially from lunges, before i could hardly walk for days, now small soreness only, mix with protien, and mango juice no prob

    For those that hate the bitter taste, please realize that this is a pre-digested (hydrolyzed) protein and the bitter taste means that it is what it's advertised to be. That's why the serving size is 10g as opposed to 50g. This is also why it's $45 per pound instead of $10-15. $45 for a 30-day supply (if taken 3x a day) is a very good price for the benefit that Leucine peptides provide rather than paying $20-30 for 30 servings of 's version of BCAA's, which haven't scientifically been proven to provide any benefits to muscle growth whatsoever.

    had this recommended for my body composition and when i still had lower leg pains from training and running. pains are almost gone now after using it daily for 10+ days. downside is it tastes terrible. i mix it with 2 packets of crystal light and water with 3 tsp of it and im able to get it down.

    Great stuff! Yeah, it tastes a little bitter, but not nearly as bad as what the previous reviews make it out to be. Stay away from artificial sweetners. If you absolutley have to have a sweetner then use Stevia, or even plain old sugar is going to be far healthier than Splenda (sucralose) or Aspartame.

    Amazing stuff. Work out super hard and notice now that I barely if at all get sore afterwards. Taking 1 tsp mixed with Hardcore Anabolic protein mix and dextrose during w/o. And yes smell and taste are horrible. Just mix it in you'll be fine.
